三种热带常见海参体腔细胞类型和数量

Types and quantities of coelomocytes in three common tropical sea cucumber species

  • 摘要: 本实验用光学显微镜对三种热带常见海参(每种n=5):黑海参(Holothuria atra)、玉足海参(Holothuria leucospilota)和红腹海参(Holothuria edulis)的体腔细胞类型及各类型细胞的相对丰度进行了研究。结果显示,三种海参的体腔细胞组成一致,均包含7大类细胞:吞噬细胞(含丝状与瓣状两种亚型)、纺锤细胞、桑葚细胞、结晶细胞、淋巴细胞(具两种形态)、血细胞及祖细胞。其中,吞噬细胞可进一步分为丝状吞噬细胞和瓣状吞噬细胞。此外,不同海参种类间同类型体腔细胞的相对丰度存在差异,红腹海参中,丝状吞噬细胞(34.63±0.38)%与桑葚细胞(19.77±0.60)%占比最高;黑海参的瓣状吞噬细胞 (27.04±0.29)%、纺锤细胞及淋巴细胞相对更为丰富;相比之下,玉足海参的结晶细胞(34.00±0.33)%与血细胞(16.30±0.57)%占比显著较高(P<0.01)。这些差异反映了不同物种在面对特定生态压力时,对细胞资源的差异化分配策略。基于COI基因的祖先状态重建分析提示,海参属核心体腔细胞谱系具有进化保守性,但在特定分支中亦存在细胞类型的特异性演化。

     

    Abstract: We examined the types of coelomocytes and their relative abundances in three common tropical sea cucumbers: Holothuria atra, Holothuria leucospilota, and Holothuria edulis (n=5 per species). The results showed that these species share the same coelomocyte types, totaling seven categories: phagocytes (subdivided into filopodial and petaloid forms), spindle cells, morula cells, crystal cells, lymphocytes (two morphotypes), hemocytes, and progenitor cells. However, significant interspecific variations were observed in the relative abundances of these cells, reflecting diverse immune strategies and ecological adaptations. Specifically, H. edulis was characterized by higher proportions of filopodial phagocytes (34.63%±0.38%) and morula cells (19.77%±0.60%); H. atra was relatively enriched in petaloid phagocytes (27.04%±0.29%), spindle cells, and lymphocytes; in contrast, H. leucospilota exhibited significantly higher proportions of crystal cells (34.00%±0.33%) and hemocytes (16.30%±0.57%). Progenitor cell abundance also varied among species, being highest in H. edulis (10.98%±0.23%), followed by H. atra (4.96%±0.23%), and lowest in H. leucospilota (3.32%±0.47%). Ancestral state reconstruction based on the COI gene suggests that while the core coelomocyte lineages are evolutionarily conserved within the genus Holothuria, specific branches exhibit lineage-specific evolution characterized by cell type heterogeneity.
