阻遏引物在入侵虎龙杂交斑食物组成分析的有效性

Effectiveness of blocking primers in the diet analysis of the invasive Hulong hybrid grouper

  • 摘要: 为了解决使用DNA宏条形码技术对具有高空胃率的入侵虎龙杂交斑胃含物残余DNA扩增过程中面临的捕食者DNA高度污染问题,设计了3条退火阻遏引物并进行测试,随后将阻遏引物应用于入侵虎龙杂交斑胃含物残余DNA分析。结果表明:具有最高解链温度(melting temperature,Tm)的阻遏引物EFELblock2在退火温度56 ℃且浓度为10 μmol·L-1时即具有最好阻断效果。添加阻遏引物不会对虎龙杂交斑潜在被捕食物种残余DNA宏条形码序列扩增造成抑制。高通量测序分析显示,添加阻遏引物后虎龙杂交斑自身序列占比显著降低且对其被捕食者多样性信息重构未产生影响。研究结果可为虎龙杂交斑风险评估提供数据支撑,也为进一步评估其对海洋生态系统的负面影响奠定重要基础。

     

    Abstract: In order to solve the high predator DNA contamination problem during the amplification of residual DNA in the stomach contents of invasive hulong hybrid grouper with high gastric emptying rate, in the report, three annealing blocking primers were designed, tested, then, the primers were used for the analysis of the residual DNA in the stomach contents of the invasive hulong hybrid grouper. The results indicated that the blocking primer EFELblock2, which has the highest Melting Temperature (Tm) value, exhibited the best blocking effect at an annealing temperature of 56 ℃ and a concentration of 10 μmol·L-1. The addition of blocking primers did not inhibit the amplification of DNA barcoding sequences from residual DNA of potential prey species in hulong hybrid grouper. High-throughput sequencing analysis results showed that the addition of blocking primers significantly reduced the proportion of self-sequences in the invasive hulong hybrid grouper and did not affect the reconstruction of prey diversity information. Our findings provided the data support for the risk assessment of hulong hybrid grouper, and laid an important foundation for further evaluating its negative impacts on marine ecosystems.
